Isolation of a sulfate ester of hyaluronic acid from heart valves.

چکیده
The most characteristic componems of the intercellular matrix, or ground substance, of connective tissue are the highly polymerized acid mucopolysaccharides. Meyer and Rapport (1) distinguish five such acid mucopolysaccharides isolated from ground substance: three chondroitin sulfates (A, B, and C), hyaluronic acid, and hyaluronosulfate. These polymers have become of particular interest in recent years as a result of increasing interest in diseases of connective tissue, commonly termed “collagen diseases,” in which the earliest and most profound alterations are found in the ground substance (2, 3). In at least one such disease, rheumatic fever, one of the most destructive lesions involves the heart valves. Meyer and Rapport (1) have reported the isolation of chondroitin sulfates B and C from heart valves and the absence of hyaluronic acid and hyaluronosulfate. The purpose of this paper is to describe the isolation from heart valves of a sulfated mucopolysaccharide acid apparently identical with hyaluronosulfate, which has been previously found only in cornea (4).

Comparative studies of the acid mucopolysaccharide composition of rheumatic and normal heart valves in man.
The nature and relative proportions of acid mucopolysacharides (AMPS) in mitral valves of normal individuals and individuals with rheumatic valvular disease were compared. The total AMPS content of valves from rheumatic patients under 21 years of age is significantly higher than that from older rheumatic patients and higher than that from age-matched normal controls. The major AMPS components o...
متن کاملIsolation and properties of hyaluronic acid from bovine heart valves.
1. A soluble extract of bovine heart valves was obtained after the tissue had been pulverized at liquid-nitrogen temperatures in a mill. 2. Hyaluronic acid was isolated from the crude extract by sedimentation equilibrium in a caesium chloride density gradient (Franek & Dunstone, 1966). 3. Analysis of the product indicated that it contained 15% of protein and the molar ratio of glucuronic acid t...
متن کاملIsolation and chemical characterization of mucopolysaccharides from rat tumors.
Mucopolysaccharides were extracted by Pronase diges tion from two types of rat tumor tissues: MC sarcoma in duced in Donryu rats by s.c. injection of methylcholanthrene and a solid tumor developed by s.c. transplantation of AH-109A cells into the animals. The mucopolysaccharides were fractionated by anion-exchange chromatography and characterized by chemical analysis, electrophoresis, infrared ...
متن کاملIsolation and Characterization of Thermophilic Alkaline Proteases Resistant to Sodium Dodecyl Sulfate and Ethylene Diamine Tetraacetic Acid from Bacillus sp. GUS1
Thermophilic Bacillus sp. GUS1, isolated from a soil sample obtained from citrus garden, produced at least three proteases as detected by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and zymogram analysis. The enzymes were stable in the alkaline pH range (8.0-12.0), with the optimum temperature and pH range of the proteases being 70ºC and 6.0-12.0, respectively. All th...
متن کاملCell-Based Microscale Isolation of Glycoaminoglycans for Glycomics Study.
Glycomics research requires the isolation of glycans from cells for structural characterization and functional studies of the glycans. A method for cell-based microscale isolation and quantification of highly sulfated, moderately sulfated, and nonsulfated glycosaminoglycans (GAGs) was developed using Chinese hamster ovary (CHO) cells. This microscale isolation relies on a mini-strong anion exch...
